γ-Gamma oryzanol significantly inhibits the activity of DNMT1 (IC50=3.2 μM), DNMT3a (IC50=22.3 μM), and DNMT3b (maximal inhibition of 57%). In contrast, the inhibitory activity of ferulic acid, a metabolite of γ-gamma oryzanol, is much lower than that of γ-oryzanol. In addition, γ-gamma oryzanol acts as a partial antagonist of ERRγ, which acts primarily as a positive regulator of DNMT1 production, thereby reducing the activity of DNMT1[1].
