WS-23, namely N,2,3-trimethyl-2-isopropylbutanamide, is a good-quality cooling agent with high cooling activity without side effects such as burning, numbness and irritation. As a cooling agent, it is mainly used in candy, oral care, and medicine.

WS-23 (FEMANO.3804) has been listed in the Chinese food additive standard GB-2760 and can be used safely. There are no reports of production and use in China. In this thesis, WS-23 and related compounds were synthesized, of which 2,3-dimethyl-2-isopropylbutyronitrile (DIPPN) is the most critical intermediate. Using propionitrile as raw material and 1,2-dimethoxyethane (DME) as solvent, DIPPN was synthesized by reaction with bromoisopropane under the action of sodium amide, with a yield of 61.5%. Under the catalysis of polyphosphoric acid (PPA), DIPPN and dimethyl carbonate (DMC) were reacted to synthesize WS-23 with a yield of 79.7%. These studies have prepared for the industrialization of WS-23. In addition, related compounds were synthesized: 2,3-dimethyl-2-isopropylbutanoic acid, N-ethyl-2,3-dimethyl-2-isopropylbutanamide, N-tert-butyl- 2,3-Dimethyl-2-isopropylbutanamide, N-p-methoxyphenyl-2,3-dimethyl-2-isopropylbutanamide. The structures of all compounds were characterized by MS, FT-IR, 1HNMR and other methods. In addition, the structure of amide compounds such as WS-23 was calculated by quantum chemical calculation method, and some information about the structure-activity relationship of cooling taste was obtained.
